アスピリンは市販薬，処方薬のいずれにも最も使われている薬剤の一つであり，臨床試験も多くなされている．急性期治療薬の使用に当たっては，薬物乱用頭痛を引き起こさないよう，使用回数を制限することも必要であるが，カフェインなどを含まない薬剤のほうが薬物乱用を引き起こしにくいとの報告もなされている．アスピリン単味，またはメトクロプラミド併用の片頭痛急性期治療の期本薬の一つとしての位置付けを検証する．

二重盲検法による比較試験（経口投与のみ）が 15 件あり．アスピリンの量はほとんどが 900-1000mg であった．アスピリン単味とプラセボとの比較ではいずれもプラセボより有意に優れていた ^{1) 2)} ．イブプロフェンとの比較 ³⁾ ではイブプロフェンが優れ，アセトアミノフェン＋コデイン ⁴⁾ ，スマトリプタン 50mg ⁵⁾ とは同等であった．薬剤の組み合わせ投与ではアスピリン（ 900mg ）＋メトクロプラミド (10mg) が最も多く ⁶⁾ ，アスピリン単味との比較では悪心の改善にやや優れていた ⁷⁾ ．スマトリプタン 100mg ^{8) 9)} ，ゾルミトリプタン 2.5mg ¹⁰⁾ との比較ではトリプタンが優れていたが，エルゴタミン＋カフェイン ¹¹⁾ との比較では，アスピリン＋メトクロプラミドが優れていた．
